Which pre-release do you have? If you installed the last pretest (2010/07/10?), i.e., the version that got frozen, then you'll be ok. If you have an earlier pretest there may be some differences in some binaries that may not get updated via TLU/tlmgr.
I'm not completely sure this is true but you could get a good idea if it's the frozen pretest by running
asy --version
in Terminal and if you have version 2.02 you might be golden.
If you are unsure install MacTeX-2010 over what you already have; it won't hurt anything.
I'd guess it will be about a week before the trickle of updates via TLU/tlmgr starts and then there should a brief rush since I've seen many updates since the freeze.
